Fabian Pfrengle, Wien: Chemical Synthesis of Plant Glycans and Their Applications in Cell Wall Biology

Plant cells are surrounded by a polysaccharide-rich matrix that constitutes the cell wall of all higher plants and provides the richest available resource of fermentable carbohydrates and bio-based materials. Optimal exploitation of this resource requires investigations into the molecular structures, interactions, and biosynthesis of cell wall glycans, which are aided by well-defined and pure glycan samples obtained through chemical synthesis. We have prepared oligo- and polysaccharides derived from different cell wall glycans using automated glycan assembly, enzymatic synthesis, and traditional solution-phase chemistry. The synthetic glycans were subsequently printed as microarrays to characterize antibodies that recognize plant cell wall glycans and glycosyltransferases that are involved in plant cell wall biosynthesis.
